Chemotherapy is the primary modality in the treatment of Nijmegen breakage syndrome. The protocol addresses dosing strategy — including the use of full-dose or a dose-modified regimen — with escalation guided by individual tolerance.
Full-dose chemotherapy is the most frequently applied method.
In previous studies, it was suggested to administer 80% of dose chemotherapy, particularly alkylating agents as cyclophosphamide, DNA topoisomerase II inhibitors, and antimetabolites such as methotrexate, and increase doses when well toleration is observed.
