Clinical scenario

Sporadic MPNST with a demonstrated BRAF V600E mutation. BRAF V600E mutations occur in a subset of sporadic MPNST cases and represent an actionable molecular target. This profile is distinct from NF1-associated MPNST and warrants a molecularly guided approach.

BRAF p(V600E) mutations are found in a subset of sporadic MPNST — the case report literature describes a single patient with MPNST with demonstrated BRAF V600E mutation treated effectively with the BRAF inhibitor vemurafenib.

There was an isolated report by Kaplan who described a female MPNST patient with a BRAF mutation who received Vemurafenib for approximately 4 days, and following this, the tumour had shrunk by 50%.
