Treatment of Advanced or Metastatic Non-Small Cell Lung Cancer with BRAF V600E Mutation
This protocol addresses a specific molecular subgroup of advanced or metastatic non-small cell lung cancer (NSCLC): patients whose tumours harbour the BRAF V600E mutation — a distinct, actionable genomic alteration.
Clinical scenario
Advanced or metastatic NSCLC in a patient with a confirmed BRAF V600E mutation. The BRAF V600E variant occurs in approximately 1%–2% of lung adenocarcinomas and represents the most common BRAF mutation in this setting.
Treatment approach
For patients with a BRAF V600E mutation who have not previously received a BRAF inhibitor, targeted combination regimens are recommended as a subsequent therapy option following disease progression.

The BRAF V600E mutation occurs in 1% to 2% of patients with lung adenocarcinoma and is the most common of the BRAF mutations.
The Panel also recommends dabrafenib/trametinib or binimetinib/encorafenib as subsequent therapy options if the patient with a BRAF V600E mutation did not previously receive a BRAF inhibitor.
