Treatment of pMMR/MSS Metastatic Colon Cancer (KRAS, NRAS, and BRAF Wild-Type) After Progression Through All Available Regimens

This protocol covers a specific and molecularly defined subset of metastatic colon adenocarcinoma: patients with proficient mismatch repair (pMMR) and microsatellite stable (MSS) disease, whose tumours carry no actionable mutations in KRAS, NRAS, or BRAF, and who have progressed through all previously available lines of treatment.

Molecular & Disease Context

Eligibility is defined by a triple wild-type, MSS biomarker profile in the setting of metastatic disease following exhaustion of prior regimens.

pMMR / MSS KRAS wild-type NRAS wild-type BRAF wild-type Metastatic Post all prior lines
Treatment Approach (Partial Overview)

For this biomarker profile after progression through all available regimens, salvage options include oral cytotoxic combinations and selective kinase inhibitors, with certain combinations preferred over monotherapy. The complete regimen choices, selection criteria, and sequencing are in the full protocol.
